Part of the extracellular matrix (ECM) family of glycoproteins, laminin is an integral part of the structural scaffolding in most tissues ¹~². In its pure form, laminin alone has proven to be sufficient in supporting embryonic stem (ES) and induced pluripotent stem (iPS) cell growth in the undifferentiated state in defined media with and without small molecule kinase inhibitors³.
Laminin was first isolated from the mouse Engelbreth-Holm-Swarm (EHS) tumor, and is the main component of the reconstituted basement membrane (RBM)⁴. In addition, laminin can serve as an alternative to RBM for the attachment of pluripotent human cell lines with or without serum, and can be used to support derivation and promote the differ -entitation of lineage-restricted progenitor cells ³,⁵~⁶.
